The overall objective of this project is to continue to enhance the capability of the Cornell University Genetically Engineered Machine (GEM) platform for the genetic modification of cell lines and primary cells. Our goal is to develop a robust pipeline for the implementation of targeted genomic editing of genes and manipulation of gene function in somatic models using GEM. As a secondary goal, in addition to improving the capabilities of our platform, we plan to develop a protocol optimized platform to efficiently create gene knockout, knockin and allele swapping cell lines.
